Due to sound hole placement, my current build has no room for pickup at neck end. What sound difference would placing it at Box center have with bridge located about 3" away compared to my planned 5"?

It’ll give you more of a midrangey sound rather than the warmer neck position. Which is not bad, think Clapton, he favors the middle pickup for his solos. Cheers 

If you have a CBG you can always try this experiment. It will give you and idea of what your pickup placement will sound like.

Wire your pickup to a volume knob.

Use a scrap piece of wood wide enough to hold your pickup. And wide enough to go across the face of your guitar. then two pieces of scrap to hold it above your strings. Painter's tape to hold it all down.

You wont be able to strum there. But you'll still be able to hear it. You could also move it around the area you have to work with. 

Hi Brent any gain in sharpness by moving pickup back a small way could be modified at the tone control on your guitar or amp.

The CBG pictured has a Gibson Les Paul pickup it produces the sharpest sound you can imagine due to is positioning, and it is a bridge position pickup. But with the use of the tone controls I can get a neck pickup warmer tone from it. Good enough for a CBG through a less than perfect amp, most of the time.
