I don't see why it wouldn't work, but you would probably have to remove the empty tuning peg or immobilize it. i've noticed empty tuners tend to vibrate when the remaining strings are played, causing a lot of buzz.Then again, and this is just me,if I had an old tenor guitar, i would probably just leave it as a four stringer. This would allow for more options. You can always tune to whatever open tuning you like and just play three of the strings without touching the other one. Sometimes i only play one string on my 3 and 4 stringers. Another issue that would have to be addressed is changing the bridge saddle locations for strings. Me, I'd leave well enough alone.

Joe is right about removing the tuner (machine head) not used.  A tenor as a  4 string will be fine.  And you can always chan ge it back.
