By twangy, do you mean a buzz? Often it can be caused by a string not well seated in the nut slot. Sometimes it's a high spot in a fret. Many tutorials on doing a "setup" online. Much help here, and lots of great free instruction on stewmac.
Might be your nut. which ever string it is see how deep it sets in the groove. You might need to un string and fill the groove in a bit and re file a wee bit. Sounds like it's hitting the fret. Good playing.
I had a bad wind where the string was coming off the tuning peg way too high, to the point where the string was barely waving at the notch on the nut. More of a "plop" than a twang, but checking the nut attack angle from a side view is my first move now.
Yes, check the bridge and especially the nut. Filing the slots in the nut is important. The slot should be deeper on the tuner side, and shaped to get the string pointing towards the tuner. Also they say the slot should not be much more deep than 1/2 the string thickness and not overly wide.
Making mistakes is what it's all about. I still have to ask for help.

Yes and with many boxes, the "back" makes a better sounding "front" anyway.
Yeah, but its not just the way its attached or secured, sometimes its better wood, different thickness, lacking any thick paper, etc. Boxes vary. A lot.
I have yet to build a box instrument without securing the lid, but I don't think I'd ever use silicone.......
Usually I try to make it so the outside looks unmodified, but the inside is reinforced with linings and blocks as needed to make it pretty durable, of course without hurting the sound potential. The boxes tend to be solid as a brick except for the top and backs ability to resonate. I tend to construct as if they were conventional instruments that look like cigar boxes, if you know what I mean. Linings, corner blocks or strips, wood glue, lid glued on.
