for some reason often my "enclosed" tuners  go out of tune, I've tightened the screw at the head and now I've stripped one... So my question is... Does tightening the screw keep the tuners and strings tight? or am I doing it wrong... and how do locking tuners work vs standard tuners?  Thanks

the one at the end of the knob that you twist to tune/tighten the string

I had problems with a few tuners- I have found that the hole you drill needs to be as perpendicular to the headstock as possible,  hope this helps,

No, that screw doesn't change the tuning, it only holds on the tuning knob. Tightening it will not make the guitar stay in tune. Try to have fewer windings on the post as well. More windings will give you more stretch and the string will take longer to get used to the tension.

Just to fulminate further on what A Russell Eight Spades CBG said:

 

3-4 wraps on a tuning post is plenty. Also, if your tuning post has a lock nut and spacer, try cranking down onthe lock nut. As Wade sez, the straighter and closer to the proper diameter the tuner post holes are, the more likely to stay in tune. I also do some string stretching when I first put a set on, by tuning the entire guitar up at least one whole note beyond where you want it to be for each string. I rarely break any doing this, and it seems to take the stretch out of the strings faster than "playing them in." 

Actually to make the most of your instrument you should give the string enough winds to be down as close to the head as you can without doubling over on itself. Keep the string under tension with your other hand as you do it so that it winds on neatly. The closer you get it to the head the better downward angle you get from your nut and better tone and sustain. Make sure those winds are neat and don't overlap! This is the biggest cause of tuning problems for beginners. 3-4 winds is plenty for the big fat E string but as you get to the lighter strings you're best to put quite a few more than that. Don't mess with those screws on the end unless you want to change out the knobs. One more thing, hopefully youre installing the correct tuners on the right side of the head, a lot of guys here get it wrong. You want the gear beyond the string post, not before, so that the string tension is pulling the post onto the gear not away from it. Good luck

I see from some of your pics that some of your machine heads are backwards, that might make them slip the gear.
